|1. Development of novel technolgies for glycomics|
We have been developong novel technologies for glycomics. One of the developed
technology is high-density lectin microarray containing 96 lectins. We
have also developed glycocomjugate microarray for high-throughput analysis
of the glycan-binding specificity of glycan-binding proteins. We are also
interested in the creation of novel probes for glycome analysis using evolutionary
|2. Glycome analysis of various cells and vesicles from bacteria to mammals|
Using the developed system, we have analyzed the glycome of various cells
derived from bacretia to mammals, including ES/iPS cells, messenchymal
stem cells, pancreatic cancer cells, lactic acid bacteria, and fungi. Recently,
we have analyzed the glycome of extracellular vesicles derived from iPS
|3. Development of the quality control system of stem cells|
Through comprehensive glycome analysis of iPS/ES cells, we have identified
a lectin called rBC2LCN, which binds specificallty to ES/iPS cells, but
not to differentiated cells. Using rBC2LCN, we have developed staining
reagents of ES/iPS cells. We have also developed a nondestructive method
to detect ES/iPS cells using cell culture media. Finally, we have developed
a reagent to specifically eliminate ES/iPS cells by fusing rBC2LCN with
cytotoxic drugs. We have also found that a2-6Sia is a marker of the differentiation
potential of messenchymal stem cells.
|4. Development of lectin-drug conjugates (LDC) for cancer therapy|
We have also found that rBC2LCN binds to pancreatic cancer stem cells.
The fusion protein of rBC2LCN with a cytotoxic drug showed strong anti-cancer
effect for pancreatic cancer mouse models. We have now challenging to apply
LDC for pancreatic cancer thrapy. We have also trying to identify novel lectins for other cancers.

We have also been developong novel diagnostic system for various diseases such as cancers and mental illnesses.
